Indications
Mécanisme d'action
Binds APRIL with a KD of 0.95 pM, blocking signaling at BCMA and TACI receptors. Inhibiting APRIL reduces serum galactose-deficient IgA1 (Gd-IgA1), which is implicated in the pathogenesis of IgAN.
Posologie adulte habituelle (résumé de la notice)
Primary IgA nephropathy at risk for progression (reduce proteinuria): 400 mg, subcutaneous injection, once every 4 weeks

- Unknown No human data. Monoclonal antibodies cross the placenta, mostly in the second and third trimesters. A monkey ePPND study showed no adverse embryofetal or postnatal effects at about 10-times clinical exposure.
- Allaitement
- Unknown No data on presence in human milk or effects on the infant or milk production. Weigh the benefits of breastfeeding against the mother's clinical need and potential adverse effects on the infant.

Contre-indications
- Serious hypersensitivity to sibeprenlimab-szsi or any excipients.
Précautions particulières
- Immunosuppression and increased infection risk: assess for active infection before starting and monitor during treatment. Patients with chronic or recurring infections may be at higher risk of serious infection.
- Limited data with concomitant systemic immunosuppressants; consider the potential for increased immunosuppression.
- Live vaccines are not recommended within 30 days before starting or during treatment; may interfere with vaccine responses.
