Gamma secretase inhibitor that blocks proteolytic activation of the Notch receptor; dysregulated Notch can activate pathways that contribute to tumor growth.

Progressing desmoid tumors requiring systemic treatment: 150 mg orally twice daily until disease progression or unacceptable toxicity.

- Unsafe Can cause fetal harm or pregnancy loss: in rats it caused embryo-fetal toxicity and death below human exposure; no human data. Check pregnancy status; use effective contraception during treatment and for 1 week after. It may weaken hormonal contraceptives, so add a barrier method.

- Unsafe No data on passage into milk or infant effects. No LactMed record. US label: do not breastfeed during treatment and for 1 week after the last dose because of possible serious infant reactions.
