Brigatinib is a tyrosine kinase inhibitor with in vitro activity against multiple kinases including ALK, ROS1, IGF-1R and FLT-3, as well as EGFR deletion and point mutations. It inhibits ALK autophosphorylation and downstream signaling (STAT3, AKT, ERK1/2, S6).

  • ILD/pneumonitis: monitor for new or worsening respiratory symptoms, especially in the first week; withhold and evaluate; discontinue permanently for Grade 3 or 4.
  • Hypertension: monitor blood pressure after 2 weeks and at least monthly; withhold, reduce or discontinue for severe cases.
  • Bradycardia: monitor heart rate and blood pressure; review concomitant bradycardic drugs.
