Inhibits histone deacetylases HDAC1, HDAC2, HDAC3 (Class I) and HDAC6 (Class II), causing accumulation of acetylated histones, open chromatin and transcriptional activation, and in vitro cell cycle arrest and/or apoptosis of some transformed cells.

  • Thromboembolism: pulmonary embolism occurred in 5% (4/86) and DVT reported; monitor for signs and symptoms.
  • Myelosuppression: dose-related thrombocytopenia and anemia; monitor blood counts every 2 weeks for the first 2 months, then monthly.
  • Gastrointestinal toxicity: nausea, vomiting, diarrhea may need antiemetics, antidiarrheals and fluid/electrolyte replacement; control pre-existing symptoms before starting.
