Summary
Dabrafenib capsules dissolve less well at higher gastric pH, so a proton pump inhibitor may lower dabrafenib absorption and potentially reduce antitumor efficacy. Dabrafenib also induces CYP3A4, which may modestly lower lansoprazole exposure.
What this interaction means in plain words
Dabrafenib and Lansoprazole can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Dabrafenib belongs to Oncology; Lansoprazole belongs to Gastrointestinal Tract.
Why it happens
Raised gastric pH reduces dabrafenib solubility, and dabrafenib induces CYP3A4 and CYP2C19, increasing lansoprazole metabolism.
What to do
Monitor tumor response and consider whether acid suppression is still needed. If reflux symptoms worsen, titrate lansoprazole dose to symptoms.
What to ask your pharmacist
- Should I take them at different times of day?
- Does the dose of either medicine need to change?
- Is there anything I should avoid, such as alcohol or certain foods, while on both?
