Summary
Combining a serotonin-norepinephrine reuptake inhibitor with an amphetamine can raise the risk of serotonin toxicity and add to cardiovascular stimulation such as raised blood pressure and heart rate. Venlafaxine may also modestly raise amphetamine exposure through CYP2D6 inhibition, although this effect is weak.
What this interaction means in plain words
Dextroamphetamine and Venlafaxine can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Dextroamphetamine belongs to Central Nervous System; Venlafaxine belongs to Central Nervous System.
Why it happens
Additive serotonergic and noradrenergic effects, with weak venlafaxine inhibition of CYP2D6-mediated amphetamine metabolism.
What to do
Monitor blood pressure, heart rate and for signs of serotonin toxicity (agitation, tremor, hyperthermia, clonus), especially after dose changes. Reduce the dose or stop one agent if symptoms appear.
What to ask your pharmacist
- Should I take them at different times of day?
- Does the dose of either medicine need to change?
- Is there anything I should avoid, such as alcohol or certain foods, while on both?
