Summary
Lefamulin inhibits CYP3A4 and itself can prolong the QT interval, so it may raise pimozide levels while adding to QT effects. This raises the risk of torsades de pointes.
What this interaction means in plain words
Lefamulin and Pimozide are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
Lefamulin belongs to Anti Bacterial; Pimozide belongs to Central Nervous System.
Why it happens
CYP3A4 inhibition reduces pimozide clearance, with additive QT prolongation.
What to do
Avoid the combination. Use an alternative antibiotic for the infection being treated.
