Summary

Both agents depress the CNS, so combined use can cause marked sedation and impaired psychomotor performance. Pentobarbital is also a CYP3A4 inducer and may lower pimozide exposure with repeated dosing, reducing antipsychotic effect.

What this interaction means in plain words

Pentobarbital and Pimozide can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.

Evidence level: Moderate

Why it happens

Additive CNS depression, plus barbiturate induction of CYP3A4-mediated pimozide metabolism.

What to do

Monitor sedation and respiratory status, and watch for loss of pimozide efficacy during prolonged barbiturate use. Adjust doses based on clinical response.

What to ask your pharmacist
