Methylene blue inhibits MAO-A and, with a serotonergic tricyclic such as amitriptyline, can precipitate serotonin syndrome. This has been reported with other serotonergic antidepressants and the risk is labelled.

Amitriptyline and Methylene blue are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
