Indications
Mécanisme d'action
HMG-CoA reductase inhibitor, inhibits rate-limiting step in cholesterol biosynthesis by competitively inhibiting HMG-CoA reductase
Posologie adulte habituelle (résumé de la notice)
Hyperlipidemia: Recommended starting dose: 2 mg PO qDay May increase to 4 mg PO qDay if necessary
Mode de prise habituel
Statins are taken by mouth once a day, long term, with blood tests to check the response and liver function.
Grossesse et allaitement
- Grossesse
- Unsafe EU labels contraindicate it in pregnancy and in women not using contraception (reduced fetal cholesterol synthesis); stop 1 month before conceiving. US label (2026) no longer does: stop when pregnancy is recognised unless the need is compelling; statin data show no rise in birth defects.
- Allaitement
- Unsafe Not known if it passes into human milk; it passes into rat milk and is 99% protein bound. EU label: contraindicated; US label: not recommended (possible effects on infant lipid metabolism). LactMed: consensus is not to use it; some experts consider pravastatin or rosuvastatin lower risk. Avoid.
Effets indésirables signalés
- Diarrhea
- Constipation
- Back pain
- Myalgia
- Pain in extremity
Précautions particulières
- Increased serum transaminases reported; typically, elevations are transient and either resolved or improved on continued therapy or after briefly interrupting therapy
- Myopathy risk factors include: age ≥65 yr, uncontrolled hypothyroidism, renal impairment, coadministration of drugs that decrease statin clearance or add to myopathy risk
- Contraindicated: Coadministration with cyclosporine
Unexplained muscle pain, tenderness or weakness while taking a statin should be reported promptly. Statins are not used in pregnancy, and some other medicines raise their levels and risk.
Informez votre pharmacien ou votre médecin en cas de grossesse ou d'allaitement, de maladie rénale ou hépatique, d'allergies, ainsi que de tout autre médicament ou complément que vous prenez.
