Összefoglaló

Butabarbital is a barbiturate that induces CYP3A4. Strong CYP3A inducers markedly lower lorlatinib exposure and caused liver enzyme elevations in a study with rifampin, so efficacy may be lost.

Mit jelent ez a kölcsönhatás közérthetően

Butabarbital and Lorlatinib are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.

Bizonyítékszint: Moderate

Miért jön létre

Induction of CYP3A4 increases lorlatinib metabolism and lowers its plasma concentrations.

Teendő

Avoid the combination and use a non-inducing sedative. Allow the inducer to wash out before starting lorlatinib.
