Per cosa si usa
Come agisce
Burosumab-twza binds to and inhibits the biological activity of FGF23, restoring renal phosphate reabsorption and increasing serum 1,25 dihydroxy vitamin D.
Dose abituale nell'adulto (sintesi del foglio illustrativo)
Adult XLH: starting 1 mg/kg rounded to nearest 10 mg, maximum 90 mg, subcutaneously every four weeks; may increase to 0.5 mg/kg (max 90 mg) every 2 weeks, then 1 mg/kg (max 90 mg) every 2 weeks if needed.
Gravidanza e allattamento
- Gravidanza
- Unknown No human data. In monkeys there were no birth defects, but fetal loss and preterm birth at high exposure with high maternal phosphate. Monitor serum phosphate throughout pregnancy.
- Allattamento
- Unknown No data on milk levels or infant effects. As a large antibody, infant absorption is likely low. Weigh breastfeeding benefits against the mother's need.
Effetti indesiderati segnalati
- Pyrexia
- Injection site reaction
- Cough
- Vomiting
- Headache
- Tooth abscess
Controindicazioni
- Concomitant use with oral phosphate and/or active vitamin D analogs, due to risk of hyperphosphatemia.
- When serum phosphorus is within or above the normal range for age.
- Severe renal impairment or end stage renal disease.
Chi deve prestare attenzione
- Hypersensitivity reactions (rash, urticaria) reported; discontinue if serious.
- Hyperphosphatemia may increase nephrocalcinosis risk; dose interruption or reduction may be required.
- Hypercalcemia: monitor calcium and PTH in high-risk patients; do not administer in moderate to severe hypercalcemia until managed.
