Come agisce

Trametinib is a reversible inhibitor of MEK1 and MEK2 activation and kinase activity. MEK proteins are upstream regulators of the ERK pathway, which promotes cellular proliferation.

Adults, all approved indications (tablets): 2 mg orally once daily, until disease progression or unacceptable toxicity; adjuvant melanoma up to 1 year

Gravidanza e allattamento

Gravidanza
Unsafe Can cause fetal harm: in rabbits it was embryotoxic and caused abortion at about 0.3 times human exposure; human data are insufficient. Check pregnancy status; contraception during treatment and for 4 months after the last dose (men with female partners: condoms for 4 months).
Allattamento
Unsafe No human milk data. Highly protein bound, but its half-life of about 4 to 5 days could let it build up in the infant. Do not breastfeed during treatment and for 4 months after the last dose.
