Summary
Vardenafil greatly enhances the vasodilator effect of nitrates such as amyl nitrite. This can cause severe, potentially fatal hypotension.
What this interaction means in plain words
Amyl nitrite and Vardenafil are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
Why it happens
Nitrates increase cGMP through nitric oxide and vardenafil inhibits its degradation by PDE5, causing excessive vasodilation.
What to do
Do not co-administer, including recreational nitrite inhalants. Keep a safe interval, at least 24 hours, before any nitrate is given after vardenafil.
