Summary

Dabrafenib solubility falls as gastric pH rises, so acid suppression may reduce its absorption. Dabrafenib also induces CYP enzymes and may lower pantoprazole exposure. Either effect may reduce efficacy of the respective drug.

What this interaction means in plain words

Dabrafenib and Pantoprazole can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.

Dabrafenib belongs to Oncology; Pantoprazole belongs to Gastrointestinal Tract.

Evidence level: Moderate

Why it happens

Higher gastric pH decreases dabrafenib dissolution, and dabrafenib induction of CYP2C19 and CYP3A4 increases pantoprazole metabolism.

What to do

Monitor tumor response and control of acid-related symptoms. Increase the pantoprazole dose if symptoms are not controlled.

What to ask your pharmacist

  • Should I take them at different times of day?
  • Does the dose of either medicine need to change?
  • Is there anything I should avoid, such as alcohol or certain foods, while on both?
