Summary

Growth hormone inhibits 11-beta-hydroxysteroid dehydrogenase type 1, which converts inactive cortisone into cortisol. Starting growth hormone can lower cortisol availability, and may reduce the effect of cortisone-type glucocorticoids that need this activation.

What this interaction means in plain words

Deflazacort and Lonapegsomatropin have a small, usually unimportant interaction. Most people can take both; the pharmacist may still mention it so that any unexpected effect is recognised.

Deflazacort belongs to Miscellaneous; Lonapegsomatropin belongs to Endocrine.

Evidence level: Low

Why it happens

Inhibition of 11-beta-HSD1 by growth hormone reduces local cortisol regeneration; deflazacort is an active-metabolite prodrug not dependent on this enzyme.

What to do

Monitor for adrenal insufficiency symptoms after starting lonapegsomatropin in patients on replacement glucocorticoids, and adjust the dose if needed.
