Encorafenib can prolong the QT interval in a dose-related way, and imipramine adds to this effect. The risk of torsades de pointes is therefore increased, particularly with electrolyte disturbance or cardiac disease.
What this interaction means in plain words
Encorafenib and Imipramine can interact in a way that may be serious. Using them together is sometimes necessary, but it needs a deliberate decision by the prescriber, a plan for monitoring, and clear advice on the warning signs to watch for.
Encorafenib belongs to Oncology; Imipramine belongs to Central Nervous System.
Why it happens

What to do
Avoid the combination if possible. Otherwise monitor ECG and electrolytes, and reduce or withhold encorafenib if QTc becomes markedly prolonged.
