Summary
Ponatinib inhibits P-glycoprotein and may modestly increase erythromycin exposure. Erythromycin, as a CYP3A4 inhibitor, may also increase ponatinib levels and its adverse effects, including hepatotoxicity, pancreatitis and cardiovascular events.
What this interaction means in plain words
Erythromycin and Ponatinib can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Erythromycin belongs to Anti Bacterial; Ponatinib belongs to Oncology.
Why it happens
Ponatinib inhibits P-glycoprotein efflux of erythromycin, and erythromycin inhibits CYP3A4 metabolism of ponatinib.
What to do
Monitor for erythromycin adverse effects and for ponatinib toxicity, including liver enzymes and lipase. Consider an alternative antibiotic that does not inhibit CYP3A4.
