What this interaction means in plain words

Lorlatinib and Ombitasvir / paritaprevir / ritonavir and dasabuvir can interact in a way that may be serious. Using them together is sometimes necessary, but it needs a deliberate decision by the prescriber, a plan for monitoring, and clear advice on the warning signs to watch for.

Evidence level: Moderate

Why it happens

Lorlatinib induces CYP3A4, increasing metabolism of the CYP3A-dependent components of the regimen.

What to do

Avoid concurrent use and select an alternative HCV regimen or ALK inhibitor. If unavoidable, monitor HCV RNA and lorlatinib toxicity including lipids and liver enzymes.
