نحوه اثر
Osimertinib is a tyrosine kinase inhibitor that irreversibly inhibits EGFRs harboring sensitising mutations and the TKI-resistance mutation T790M.

EGFR mutation-positive NSCLC (monotherapy): 80 mg osimertinib orally once a day; adjuvant until recurrence or unacceptable toxicity (not studied beyond 3 years); locally advanced or metastatic until progression or unacceptable toxicity

- Unsafe No human data. In rats it caused embryo loss, reduced fetal growth and newborn deaths at about 1.5 times human exposure. Avoid unless the woman's condition requires it; contraception for 6 weeks (US) or 2 months (EU) after the last dose; male patients 4 months.
- شیردهی
- Unsafe No milk data; its long half-life (about 48 h) and active metabolites may build up in the infant, and rat pups had poor growth and deaths. Do not breastfeed during treatment and for 2 weeks after the last dose (US); EU: stop breastfeeding.

موارد منع مصرف
- Hypersensitivity to the active substance or any excipient.
- St. John's Wort must not be used together with osimertinib.

- Severe, life-threatening or fatal ILD/pneumonitis; interrupt treatment for acute or worsening pulmonary symptoms and discontinue if ILD is diagnosed.
- QTc prolongation: avoid in congenital long QT syndrome when possible; consider periodic ECG and electrolyte monitoring.
- Decreased LVEF: consider cardiac monitoring in patients with cardiac risk factors.
