Mécanisme d'action
Kinase inhibitor that binds to and inhibits enzymatic activity of FGFR1, FGFR2, FGFR3 and FGFR4 (in vitro data); inhibits FGFR phosphorylation and signaling and decreases viability of cell lines with FGFR alterations.
Posologie adulte habituelle (résumé de la notice)
Locally advanced or metastatic urothelial carcinoma with susceptible FGFR3 alterations: 8 mg orally once daily; increase to 9 mg once daily based on tolerability, including hyperphosphatemia, at 14 to 21 days; continue until disease progression or unacceptable toxicity

- Unsafe Can cause fetal harm: in rats it caused malformations and embryo-fetal death below human exposure. No human data. Verify pregnancy status; women, and men with female partners, use effective contraception during and for 1 month after.
- Allaitement
- Unsafe No milk data. Because of possible serious reactions in the infant, do not breastfeed during treatment and for 1 month after the last dose.

Contre-indications
- None.
Précautions particulières
- Ocular disorders including CSR/RPED (22%) and dry eye; monthly eye exams for first 4 months then every 3 months, and urgently for visual symptoms; dry eye prophylaxis.
- Hyperphosphatemia and soft tissue mineralization; monitor phosphate, restrict dietary phosphate to 600–800 mg daily, consider phosphate binder if above 7.0 mg/dL.
- Embryo-fetal toxicity; verify pregnancy status and use effective contraception during treatment and for one month after last dose.
