Indications

Mécanisme d'action

Givinostat is a histone deacetylase inhibitor. The precise mechanism by which it acts in DMD is unknown.

Grossesse et allaitement

Grossesse
Unsafe Used for Duchenne muscular dystrophy, which affects almost only boys, so there are no pregnancy data. In rats and rabbits it lowered fetal weight and raised fetal and pup deaths and behaviour changes, some below human exposure.
Allaitement
Unknown No human or animal data on milk. LactMed: highly protein-bound, so milk levels are likely very low; not a reason to stop breastfeeding, but use with caution and watch the infant for diarrhoea, vomiting, bruising or bleeding.

Effets indésirables signalés

  • Diarrhea
  • Abdominal pain
  • Thrombocytopenia
  • Nausea/vomiting
  • Hypertriglyceridemia
  • Pyrexia

Précautions particulières

  • Dose-related thrombocytopenia and myelosuppression (anemia, neutropenia); monitor blood counts every 2 weeks for first 2 months, at month 3, then every 3 months.
  • Increased triglycerides; monitor at 1 month, 3 months, 6 months, then every 6 months. Discontinue if they remain elevated despite diet and dose adjustment.
  • Gastrointestinal disturbances (diarrhea, nausea/vomiting, abdominal pain); consider antiemetics or antidiarrheals and replace fluids and electrolytes.

Informez votre pharmacien ou votre médecin en cas de grossesse ou d'allaitement, de maladie rénale ou hépatique, d'allergies, ainsi que de tout autre médicament ou complément que vous prenez.
