Indications
Mécanisme d'action
Olaparib inhibits PARP enzymes (PARP1, PARP2, PARP3). Cytotoxicity may involve inhibition of PARP enzymatic activity and increased formation of PARP-DNA complexes, causing DNA damage and cancer cell death, with greater activity in cells deficient in BRCA1/2, ATM or other HRR genes.
Posologie adulte habituelle (résumé de la notice)
All indications: 300 mg orally twice daily, with or without food; swallow tablets whole, do not chew, crush, dissolve or divide
Grossesse et allaitement
- Grossesse
- Unsafe No human data. Based on its mechanism (PARP inhibition) and rat studies showing malformations and embryo loss below human exposure, it can cause fetal harm. Avoid; confirm a negative pregnancy test, and use effective contraception during treatment and for 6 months after (EU: two methods).
- Allaitement
- Unsafe No data on milk levels or infant effects. Because of possible serious reactions in the infant, do not breastfeed during treatment and for 1 month after the last dose (US label); the EU label contraindicates breastfeeding.
Effets indésirables signalés

- Fatigue
