Résumé
Esomeprazole can substantially lower absorption of acalabrutinib tablets or capsules, which may weaken its anticancer effect. Because PPI acid suppression lasts a long time, separating the doses does not reliably prevent this.
Ce que signifie cette interaction, en termes simples
Acalabrutinib and Esomeprazole can interact in a way that may be serious. Using them together is sometimes necessary, but it needs a deliberate decision by the prescriber, a plan for monitoring, and clear advice on the warning signs to watch for.
Acalabrutinib belongs to Oncology; Esomeprazole belongs to Gastrointestinal Tract.
Pourquoi elle survient
Higher gastric pH reduces the pH-dependent solubility and absorption of acalabrutinib.
Conduite à tenir
Avoid PPIs with acalabrutinib. If acid suppression is needed, use an antacid taken 2 hours apart or an H2 blocker with acalabrutinib taken 2 hours before it, or switch to the tablet form, which is less affected.
