Résumé
Acalabrutinib capsules need an acidic stomach to dissolve, and lansoprazole raises gastric pH for a prolonged period. This can substantially lower acalabrutinib absorption and may reduce its anticancer efficacy.
Ce que signifie cette interaction, en termes simples
Acalabrutinib and Lansoprazole can interact in a way that may be serious. Using them together is sometimes necessary, but it needs a deliberate decision by the prescriber, a plan for monitoring, and clear advice on the warning signs to watch for.
Acalabrutinib belongs to Oncology; Lansoprazole belongs to Gastrointestinal Tract.
Pourquoi elle survient
PPI-induced rise in gastric pH reduces the solubility and absorption of acalabrutinib capsules.
Conduite à tenir
Avoid the PPI where possible; an antacid taken 2 hours apart or an H2 blocker taken 2 hours after acalabrutinib is an alternative. The tablet formulation, which is not pH dependent, can be used if a PPI is required.
