Résumé
Clarithromycin, a strong CYP3A4 inhibitor, markedly increases cobimetinib exposure and may raise the risk of adverse effects such as hemorrhage, serous retinopathy, rhabdomyolysis, and cardiomyopathy.
Ce que signifie cette interaction, en termes simples
Clarithromycin and Cobimetinib can interact in a way that may be serious. Using them together is sometimes necessary, but it needs a deliberate decision by the prescriber, a plan for monitoring, and clear advice on the warning signs to watch for.
Clarithromycin belongs to Anti Bacterial; Cobimetinib belongs to Oncology.
Pourquoi elle survient
Clarithromycin inhibits CYP3A4-mediated metabolism of cobimetinib, greatly increasing its exposure.
Conduite à tenir
Avoid the combination and use a non-CYP3A4-inhibiting antibiotic such as azithromycin. If a strong inhibitor is short-term unavoidable, reduce cobimetinib dose per labeling and monitor closely.
