Ribociclib is a moderate CYP3A4 inhibitor and also prolongs the QT interval in a dose-dependent manner. Combined with cisapride, which is a CYP3A4 substrate with a known risk of torsades de pointes, this could produce serious arrhythmia.

Cisapride and Ribociclib are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.
