Per cosa si usa
Come agisce
Linaclotide and its active metabolite are guanylate cyclase-C agonists acting locally on the intestinal epithelium. Raised cGMP stimulates chloride and bicarbonate secretion via CFTR, increasing intestinal fluid and accelerating transit.
Dose abituale nell'adulto (sintesi del foglio illustrativo)
IBS-C: 290 mcg orally once daily
Gravidanza e allattamento
- Gravidanza
- Unknown Negligibly absorbed systemically; maternal use not expected to result in fetal exposure. Available human data insufficient to inform drug-associated risk.
- Allattamento
- Safe Linaclotide and its active metabolite were not detected in milk; exposure of breastfed infants not expected. Consider benefits of breastfeeding with maternal need.
Effetti indesiderati segnalati
- Diarrhea
- Abdominal pain
- Flatulence
- Abdominal distension
- Viral gastroenteritis
- Headache
Avvertenze in evidenza
- Risk of serious dehydration in pediatric patients less than 2 years of age. Contraindicated in this age group; in neonatal mice a single clinically relevant adult oral dose caused deaths due to dehydration.
Controindicazioni
- Patients less than 2 years of age due to risk of serious dehydration.
- Known or suspected mechanical gastrointestinal obstruction.
Chi deve prestare attenzione
- Severe diarrhea may occur; post-marketing cases with dizziness, syncope, hypotension and electrolyte abnormalities (hypokalemia, hyponatremia) required hospitalization or IV fluids.
- Insufficient data on GC-C intestinal expression in children under 2 years to assess risk of diarrhea and its serious consequences.
