Come agisce
Inhibits histone deacetylases HDAC1, HDAC2, HDAC3 (Class I) and HDAC6 (Class II), causing accumulation of acetylated histones, open chromatin and transcriptional activation, and in vitro cell cycle arrest and/or apoptosis of some transformed cells.
Dose abituale nell'adulto (sintesi del foglio illustrativo)
Cutaneous T-cell lymphoma (cutaneous manifestations, after two systemic therapies): 400 mg orally once daily with food; continue as long as no progressive disease or unacceptable toxicity
Gravidanza e allattamento
- Gravidanza
- Unsafe Can cause fetal harm: an HDAC inhibitor that crosses the placenta; in rats and rabbits it harmed development at about half the human exposure. Avoid; pregnancy test within 7 days before starting; contraception during and for 6 months after (men with female partners: 3 months).
- Allattamento
- Unsafe No data on passage into milk or infant effects. No LactMed record. Because of possible serious reactions in the infant, do not breastfeed during treatment and for at least 1 week after the last dose.

Chi deve prestare attenzione
- Thromboembolism: pulmonary embolism occurred in 5% (4/86) and DVT reported; monitor for signs and symptoms.
- Myelosuppression: dose-related thrombocytopenia and anemia; monitor blood counts every 2 weeks for the first 2 months, then monthly.
- Gastrointestinal toxicity: nausea, vomiting, diarrhea may need antiemetics, antidiarrheals and fluid/electrolyte replacement; control pre-existing symptoms before starting.
