Sintesi
Encorafenib causes dose-dependent QTc prolongation, and anagrelide has a similar effect, so use together may raise the risk of torsades de pointes. The risk is greater with electrolyte abnormalities or underlying cardiac disease.
Cosa significa questa interazione in parole semplici
Anagrelide and Encorafenib can interact in a way that may be serious. Using them together is sometimes necessary, but it needs a deliberate decision by the prescriber, a plan for monitoring, and clear advice on the warning signs to watch for.
Anagrelide belongs to Cardiovascular; Encorafenib belongs to Oncology.
Perché avviene
Additive pharmacodynamic prolongation of the QT interval.
Cosa fare
Avoid the combination if possible. If used together, monitor ECG and electrolytes and reduce or interrupt encorafenib per labeling if QTc becomes markedly prolonged.
