Sintesi
Ivosidenib is associated with QT prolongation, and anagrelide has a similar effect, so the combination may increase the risk of torsades de pointes. The legacy description of a QT decrease is not consistent with the known effects of either drug.
Cosa significa questa interazione in parole semplici
Anagrelide and Ivosidenib can interact in a way that may be serious. Using them together is sometimes necessary, but it needs a deliberate decision by the prescriber, a plan for monitoring, and clear advice on the warning signs to watch for.
Anagrelide belongs to Cardiovascular; Ivosidenib belongs to Oncology.
Perché avviene
Additive pharmacodynamic QT prolongation; ivosidenib is also a CYP3A4 inducer, which may lower anagrelide exposure.
Cosa fare
Avoid the combination if possible. If used together, monitor ECG and electrolytes regularly and reduce or stop therapy if QTc becomes markedly prolonged.
