作用のしくみ

Vonoprazan suppresses basal and stimulated gastric acid secretion by inhibiting the H+,K+-ATPase enzyme system in parietal cells in a potassium-competitive manner. It binds to active pumps noncovalently and reversibly and does not require activation by acid.

  • Known hypersensitivity to vonoprazan or any component of the product (reactions have included anaphylactic shock)
  • Concomitant use with rilpivirine-containing products

  • Symptomatic response does not preclude gastric malignancy; consider further follow-up and testing, and endoscopy in older patients.
  • Acute tubulointerstitial nephritis: discontinue and evaluate if suspected.
  • May increase risk of Clostridioides difficile-associated diarrhea; use the shortest duration appropriate.

People taking proton pump inhibitors for a long time should have the need for treatment reviewed, because long-term use is linked to low magnesium, vitamin B12 deficiency, bone fractures and gut infections. Tell your pharmacist about all other medicines, since some interact.
