Summary

Vandetanib inhibits the renal uptake transporter OCT2, which helps eliminate amantadine. Amantadine exposure may therefore increase, with a risk of CNS toxicity. Vandetanib also prolongs the QT interval, and amantadine has been linked to QT effects.

What this interaction means in plain words

Amantadine and Vandetanib can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.

Amantadine belongs to Anti Virals; Vandetanib belongs to Oncology.

Evidence level: Low

Why it happens

Inhibition of OCT2-mediated renal secretion of amantadine.

What to do

Monitor for amantadine toxicity and consider a lower dose. Obtain an ECG and check electrolytes given the QT risk with vandetanib.
