Summary

Ritonavir-boosted paritaprevir inhibits CYP3A4 and raises amlodipine exposure, increasing the risk of hypotension and peripheral edema. Valsartan exposure may also increase through inhibition of hepatic transporters.

What this interaction means in plain words

Amlodipine-hydrochlorothiazide-valsartan and Ombitasvir / paritaprevir / ritonavir and dasabuvir can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.

Evidence level: Moderate

Why it happens

CYP3A4 inhibition by ritonavir reduces amlodipine clearance, and transporter inhibition (OATP/MRP2) may raise valsartan levels.

What to do

Reduce the amlodipine dose by about half, or use a different antihypertensive, and monitor for hypotension and edema. Monitor blood pressure and renal function for valsartan-related effects.

What to ask your pharmacist
