Summary
Erythromycin is a P-glycoprotein inhibitor and can raise berotralstat exposure. Berotralstat in turn inhibits P-gp and may increase exposure to P-gp substrates. Higher berotralstat levels raise the risk of gastrointestinal effects and QT prolongation.
What this interaction means in plain words
Berotralstat and Erythromycin can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Berotralstat belongs to Miscellaneous; Erythromycin belongs to Anti Bacterial.
Why it happens
Mutual P-glycoprotein inhibition reduces efflux of berotralstat and may raise erythromycin levels.
What to do
Monitor for berotralstat adverse effects such as gastrointestinal upset and consider ECG monitoring if QT risk factors exist. Consider an alternative antibiotic if prolonged therapy is needed.
What to ask your pharmacist
- Should I take them at different times of day?
- Does the dose of either medicine need to change?
- Is there anything I should avoid, such as alcohol or certain foods, while on both?
