Summary
Fexinidazole inhibits CYP3A4, and MMAE, the cytotoxic component of brentuximab vedotin, is metabolized by CYP3A4. Increased MMAE exposure may increase neutropenia and neuropathy. Fexinidazole is used only for short courses for trypanosomiasis, so the overlap would be brief.
What this interaction means in plain words
Brentuximab and Fexinidazole can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Why it happens
CYP3A4 inhibition by fexinidazole may decrease MMAE clearance.
What to do
Avoid if possible and defer brentuximab if the fexinidazole course is short. If both are needed, monitor blood counts and neuropathy.
What to ask your pharmacist
- Should I take them at different times of day?
- Does the dose of either medicine need to change?
