Summary

Celecoxib is a moderate CYP2D6 inhibitor and could raise exposure to lofepramine and its active metabolite desipramine, which are CYP2D6 substrates. This may increase anticholinergic effects, sedation or cardiac conduction effects, though clinical reports are limited.

What this interaction means in plain words

Celecoxib and Lofepramine can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.

Evidence level: Low

Why it happens

Celecoxib inhibits CYP2D6, reducing the metabolism of tricyclic antidepressants such as lofepramine and its metabolite desipramine.

What to do

Monitor for tricyclic adverse effects such as dry mouth, tachycardia and drowsiness, and consider a lower lofepramine dose if they appear. ECG monitoring is reasonable in patients with cardiac risk factors.

What to ask your pharmacist

  • Should I take them at different times of day?
  • Does the dose of either medicine need to change?
  • Is there anything I should avoid, such as alcohol or certain foods, while on both?
