Summary
Erdafitinib is metabolized by CYP2C9 and CYP3A4, and inhibition of these pathways can raise its exposure. This may increase hyperphosphatemia, ocular and dermatologic toxicity, and stomatitis.
What this interaction means in plain words
Chloramphenicol and Erdafitinib can interact in a way that may be serious. Using them together is sometimes necessary, but it needs a deliberate decision by the prescriber, a plan for monitoring, and clear advice on the warning signs to watch for.
Chloramphenicol belongs to Anti Bacterial; Erdafitinib belongs to Oncology.
Why it happens
Chloramphenicol inhibits CYP3A4 and CYP2C9, reducing erdafitinib clearance.
What to do
Use an alternative antibiotic if possible. If coadministered, monitor closely for adverse reactions and consider reducing the erdafitinib dose, and re-escalate after the inhibitor is stopped if no toxicity.
What to ask your pharmacist
- Does the benefit of taking both outweigh the risk in my case?
- Do I need any blood test, ECG or other check while on both?
- What early warning signs should I watch for, and what should I do if they appear?
