Summary

Growth hormone inhibits 11-beta-hydroxysteroid dehydrogenase type 1, which reduces conversion of cortisone to cortisol, and this can reduce the effect of cortisone-type glucocorticoids. Prednisone itself needs conversion to prednisolone by the same enzyme, so its effect may be reduced. Patients on glucocorticoid replacement may need higher doses after starting growth hormone.

What this interaction means in plain words

Lonapegsomatropin and Prednisone can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.

Lonapegsomatropin belongs to Endocrine; Prednisone belongs to Endocrine.

Evidence level: Moderate

Why it happens

Growth hormone inhibits 11-beta-HSD1, reducing activation of prednisone to prednisolone.

What to do

Monitor for signs of glucocorticoid insufficiency after starting lonapegsomatropin and increase the glucocorticoid dose if needed. Consider prednisolone or hydrocortisone-based regimens in adrenal insufficiency and reassess when GH dosing changes.
