Summary

Mifepristone antagonizes glucocorticoid receptors and can reduce the effect of corticosteroids. In patients on long-term steroid treatment this may cause adrenal insufficiency or loss of disease control, and labeling lists chronic corticosteroid therapy as a contraindication.

What this interaction means in plain words

Mifepristone and Prednisolone are not normally used together. Taking them at the same time is expected to cause harm or make one of them unsafe, so a different medicine is usually chosen for one of the two.

Mifepristone belongs to Endocrine; Prednisolone belongs to Endocrine.

Evidence level: Moderate

Why it happens

Mifepristone blocks glucocorticoid receptors, opposing the effect of prednisolone.

What to do

Do not use mifepristone in patients on long-term corticosteroid therapy. Seek specialist advice if both are needed, as standard steroid doses may be ineffective.
