Summary
Nefazodone inhibits serotonin reuptake and blocks 5-HT2A receptors, and with sumatriptan it may rarely cause serotonin syndrome. Nefazodone also inhibits CYP3A4, though sumatriptan is mainly cleared by MAO-A so this has little effect.
What this interaction means in plain words
Nefazodone and Sumatriptan can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Why it happens
Additive serotonergic effects from reuptake inhibition and 5-HT1 agonism.
What to do
Monitor for signs of serotonin syndrome, especially during initiation or dose increases. Discontinue the agents if toxicity develops.
