Severity: Moderate

Nilotinib and Pantoprazole: moderate interaction.

For healthcare professionals — reference only. This page is a brief summary, not medical advice and not a substitute for official labeling or clinical judgement. Always verify before prescribing or dispensing.

Summary

Acid suppression with a proton pump inhibitor can lower nilotinib absorption because the drug dissolves poorly at higher gastric pH. This may reduce exposure and lead to loss of antileukemic efficacy. Separating the doses does not reliably prevent the effect because of the long duration of acid suppression.

What this interaction means in plain words

Nilotinib and Pantoprazole can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.

Nilotinib belongs to Oncology; Pantoprazole belongs to Gastrointestinal Tract.

Evidence level: Moderate

Why it happens

Increased gastric pH reduces the pH-dependent solubility and absorption of nilotinib.

What to do

Avoid long-term PPI use where possible; if acid suppression is required, use a short-acting antacid or an H2 blocker timed away from nilotinib. Monitor molecular response to treatment.

What to ask your pharmacist

  • Should I take them at different times of day?
  • Does the dose of either medicine need to change?
  • Is there anything I should avoid, such as alcohol or certain foods, while on both?
