Summary

Parecoxib is converted to valdecoxib, an acidic compound that may compete with probenecid for renal tubular secretion. Probenecid can in turn raise valdecoxib exposure. The clinical effect is generally modest but may increase NSAID-related adverse effects.

What this interaction means in plain words

Parecoxib and Probenecid have a small, usually unimportant interaction. Most people can take both; the pharmacist may still mention it so that any unexpected effect is recognised.

Evidence level: Low

Why it happens

Probenecid inhibits renal organic anion transporters, reducing tubular secretion of valdecoxib, the active metabolite of parecoxib.

What to do

No routine dose change is needed; watch for signs of increased COX-2 inhibitor effects such as edema, hypertension or renal impairment during prolonged co-use.

What to ask your pharmacist

  • Is there anything I should notice while taking both?
  • Do I need to tell my doctor at the next visit?
