Mécanisme d'action

Anthracycline; intercalates between DNA base pairs, impairs topoisomerase II function and subsequently inhibits DNA and RNA replication. Doxorubicin is a strong chelator; doxorubicin-iron complex binds to cell membranes and DNA and produces free hydroxyl radicals that cleaves them.

Posologie adulte habituelle (résumé de la notice)

Grossesse et allaitement

Grossesse
Unsafe Can cause fetal harm: embryotoxic in rats and abortifacient in rabbits at low doses. Avoid in the 1st trimester; human data for doxorubicin later in pregnancy do not establish the risk, and any use needs specialist care. Use effective contraception during and for 6 months after treatment.
Allaitement
Unsafe Not known if the liposomal form passes into milk; conventional doxorubicin and its active metabolite reach high, persistent levels. Because of possible serious infant reactions, do not breastfeed during treatment. LactMed: breastfeeding is generally contraindicated with anthracyclines.
