Résumé
Etrasimod is cleared by CYP2C8, CYP2C9 and CYP3A4. Amiodarone inhibits CYP2C9 and CYP3A4 and may raise etrasimod exposure. Both drugs can slow heart rate and conduction, so bradyarrhythmia risk is a concern.
Ce que signifie cette interaction, en termes simples
Amiodarone and Etrasimod can interact in a way that may be serious. Using them together is sometimes necessary, but it needs a deliberate decision by the prescriber, a plan for monitoring, and clear advice on the warning signs to watch for.
Amiodarone belongs to Cardiovascular; Etrasimod belongs to Gastrointestinal Tract.
Pourquoi elle survient
Inhibition of CYP2C9 and CYP3A4 by amiodarone raises etrasimod levels, with additive bradycardic effects.
Conduite à tenir
Avoid the combination; labeling advises against etrasimod with moderate-to-strong dual CYP2C9/CYP3A4 inhibitors. If unavoidable, monitor heart rate and ECG.
