Sintesi
Both drugs can prolong the QT interval, and pasireotide may also cause bradycardia, which increases the risk of torsades de pointes. The risk is higher with hypokalemia, hypomagnesemia or underlying heart disease.
Cosa significa questa interazione in parole semplici
Amitriptyline and Pasireotide can interact in a way that may be serious. Using them together is sometimes necessary, but it needs a deliberate decision by the prescriber, a plan for monitoring, and clear advice on the warning signs to watch for.
Amitriptyline belongs to Central Nervous System; Pasireotide belongs to Endocrine.
Perché avviene
Additive prolongation of cardiac repolarization, with pasireotide-related bradycardia increasing the arrhythmic risk.
Cosa fare
Obtain a baseline ECG and monitor it during treatment, and correct potassium and magnesium abnormalities before and during therapy. Consider an alternative to amitriptyline if QTc becomes prolonged.
