Resumo
Mavacamten is cleared mainly by CYP2C19 with CYP3A4 contribution, and exposure changes can cause loss of efficacy or excessive negative inotropy leading to systolic heart failure. Modafinil has mixed effects, inducing CYP3A4 and inhibiting CYP2C19, making the net exposure unpredictable.
