Как действует
Edited autologous CD34+ cells engraft in bone marrow and differentiate into erythroid cells with reduced BCL11A expression, increasing gamma-globin and HbF production. In SCD, HbF reduces intracellular HbS and red cell sickling; in TDT it improves globin imbalance and reduces transfusion dependence.
Обычная доза для взрослых (кратко по инструкции)
SCD with recurrent VOCs or transfusion-dependent beta-thalassemia: minimum dose 3 × 10 6 CD34+ cells/kg, single one-time intravenous infusion; administer all vials, 48 hours to 7 days after the last myeloablative conditioning dose

- Unsafe No clinical or animal data. Must not be administered during pregnancy because of risks of myeloablative conditioning.

- Unsafe No data on milk presence or effects. Breastfeeding should be discontinued during conditioning because of myeloablative risks.

- Risk of neutrophil engraftment failure; monitor ANC and give rescue CD34+ cells if it occurs. G-CSF not recommended for 21 days after infusion.
- Delayed platelet engraftment with increased bleeding risk; monitor platelets frequently until recovery.
- Hypersensitivity including anaphylaxis can occur due to DMSO or dextran 40 in the cryopreservation solution.
