Summary
Amantadine is eliminated largely unchanged by the kidney through active tubular secretion. Trilaciclib inhibits renal cation transporters, which may raise amantadine exposure and increase the risk of neurotoxicity such as confusion, hallucinations, myoclonus and seizures.
What this interaction means in plain words
Amantadine and Trilaciclib can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Amantadine belongs to Anti Virals; Trilaciclib belongs to Oncology.
Why it happens
Trilaciclib inhibits OCT2, MATE1 and MATE2-K, reducing renal tubular secretion of amantadine.
What to do
Monitor for amantadine adverse effects such as confusion, hallucinations and myoclonus during trilaciclib dosing, and reduce the amantadine dose if they occur. Take extra care in patients with renal impairment.
What to ask your pharmacist
- Should I take them at different times of day?
- Does the dose of either medicine need to change?
- Is there anything I should avoid, such as alcohol or certain foods, while on both?
