Summary
Erdafitinib inhibits P-glycoprotein, and brentuximab vedotin's cytotoxic component MMAE is a P-gp substrate. Higher MMAE exposure could increase toxicities such as neutropenia and peripheral neuropathy. The clinical significance has not been well studied.
What this interaction means in plain words
Brentuximab and Erdafitinib can affect each other. The combination is often manageable, but it may need a change of dose, a change of timing, or extra checks while both are taken.
Brentuximab belongs to Oncology; Erdafitinib belongs to Oncology.
Why it happens
Erdafitinib inhibits P-gp efflux, potentially raising exposure to MMAE released from brentuximab vedotin.
What to do
Monitor blood counts and for peripheral neuropathy and infection. Consider dose modification of brentuximab vedotin if toxicity appears.
